(Image : Unsplash)<\/figcaption><\/figure>\n<p>L'\u00e9pig\u00e9n\u00e9tique \u00e9tudie les modifications h\u00e9r\u00e9ditaires de l'activit\u00e9 des g\u00e8nes qui ne sont pas dues \u00e0 des changements de la s\u00e9quence d'ADN primaire. \"Alors que chez les mammif\u00e8res, les marques \u00e9pig\u00e9n\u00e9tiques sont normalement r\u00e9initialis\u00e9es \u00e0 chaque g\u00e9n\u00e9ration, ce n'est pas toujours le cas chez les plantes. Chez les plantes, les modifications \u00e9pig\u00e9n\u00e9tiques au sein d'une g\u00e9n\u00e9ration peuvent \u00eatre transmises de mani\u00e8re stable \u00e0 la g\u00e9n\u00e9ration suivante et m\u00eame \u00eatre h\u00e9rit\u00e9es sur de nombreuses g\u00e9n\u00e9rations\", d\u00e9clare <a href=\"\/\/www.professoren.tum.de\/johannes-frank\">Frank Johannes, professeur d'\u00e9pig\u00e9n\u00e9tique des populations et d'\u00e9pig\u00e9nomique \u00e0 l'Universit\u00e9 technique de Munich (TUM)<\/a>.<\/p>\n<p>Son \u00e9quipe de recherche s'int\u00e9resse \u00e0 la fr\u00e9quence des \u00e9pimutations dans les g\u00e9nomes des plantes, \u00e0 leur stabilit\u00e9 au fil des g\u00e9n\u00e9rations et \u00e0 la question de savoir si elles peuvent influencer des caract\u00e9ristiques importantes des plantes.<\/p>\n<p><strong>Les arbres portent en eux de nombreuses \u00e9pimutations<\/strong><\/p>\n<p>\"En raison de leur long\u00e9vit\u00e9, les arbres peuvent nous fournir des connaissances \u00e9pig\u00e9n\u00e9tiques sur de longues p\u00e9riodes\", d\u00e9clare <a href=\"\/\/www.ias.tum.de\/active-fellows\/johannes-frank\/\">Prof. Johannes<\/a>. Avec <a href=\"\/\/www.ias.tum.de\/active-fellows\/schmitz-robert-j\/\">Robert J. Schmitz<\/a>Le professeur Hans Fisher, professeur \u00e0 l'universit\u00e9 de G\u00e9orgie (\u00c9tats-Unis) et Hans Fisher Fellow \u00e0 l'Institute for Advanced Study de la TUM (TUM-IAS), vient de publier deux \u00e9tudes \u00e0 ce sujet.<\/p>\n<p>L'\u00e9quipe s'est concentr\u00e9e sur un peuplier \u00e2g\u00e9 de 330 ans. Elle a compar\u00e9 la m\u00e9thylation de l'ADN, un m\u00e9canisme chez les plantes qui repr\u00e9sente une modification chimique de l'ADN, sur les feuilles de diff\u00e9rentes branches de l'arbre. Ils ont ainsi pu montrer que les modifications \u00e9pig\u00e9n\u00e9tiques augmentent continuellement en fonction de l'\u00e2ge de l'arbre. Plus deux feuilles \u00e9taient \u00e9loign\u00e9es l'une de l'autre par rapport \u00e0 l'\u00e2ge de d\u00e9veloppement de la branche, plus elles \u00e9taient dissemblables au niveau de la m\u00e9thylation de l'ADN. Les chercheurs en ont conclu que le taux d'\u00e9pimutations par an est environ 10.000 fois plus \u00e9lev\u00e9 que le taux de mutations g\u00e9n\u00e9tiques sur le m\u00eame arbre.<\/p>\n<p><strong>L'\u00e2ge des arbres peut \u00eatre dat\u00e9 gr\u00e2ce \u00e0 l'\u00e9pig\u00e9n\u00e9tique<\/strong><\/p>\n<p>Cette d\u00e9couverte a permis de comprendre que les \u00e9pimutations pouvaient \u00e9galement servir d'horloge mol\u00e9culaire pour d\u00e9terminer l'\u00e2ge de l'arbre. \"Seules quelques branches avaient \u00e9t\u00e9 dat\u00e9es par le comptage des cernes, mais malheureusement pas le tronc principal. Or, nous avions besoin de cette information pour notre analyse. Nous avons donc trait\u00e9 l'\u00e2ge total de l'arbre comme un param\u00e8tre inconnu et nous avons demand\u00e9 aux donn\u00e9es de m\u00e9thylation de l'ADN des feuilles de nous dire quel \u00e2ge avait l'arbre. Cela nous a donn\u00e9 une estimation d'environ 330 ans\", rapporte le professeur Johannes.<\/p>\n<p>Il s'est av\u00e9r\u00e9 par la suite que cette estimation correspondait \u00e0 la datation du tronc principal bas\u00e9e sur le diam\u00e8tre et \u00e0 d'autres informations sur l'histoire de la vie de cet arbre particulier. \"C'\u00e9tait la premi\u00e8re indication qu'il y avait une sorte d'horloge \u00e9pig\u00e9n\u00e9tique dans les arbres\".<\/p>\n<p><strong>Une fen\u00eatre sur le pass\u00e9<\/strong><\/p>\n<p>L'\u00e9quipe du professeur Johannes cherche \u00e0 pr\u00e9sent \u00e0 savoir si les changements environnementaux subis par les arbres au cours de leur longue vie laissent des signatures \u00e9pig\u00e9n\u00e9tiques qui peuvent \u00eatre lues pour en apprendre plus sur leur pass\u00e9.<\/p>\n<p>\"Notre objectif est de combiner les donn\u00e9es environnementales historiques avec notre travail \u00e9pig\u00e9n\u00e9tique afin de comprendre si les arbres 'enregistrent' \u00e9pig\u00e9n\u00e9tiquement des d\u00e9fis environnementaux sp\u00e9cifiques tels que les s\u00e9cheresses ou les variations de temp\u00e9rature.
