Il grafene ha molte propriet\u00e0 straordinarie, che rendono possibili applicazioni rivoluzionarie nei settori pi\u00f9 diversi. Non per niente l'UE ha lanciato la \"Graphene Flagship\", che \u00e8 sostenuta con un miliardo di euro ed \u00e8 quindi considerata la pi\u00f9 grande iniziativa di ricerca europea. Nell'ambito di questo enorme progetto, l'Empa contribuisce anche con il suo know-how, poich\u00e9 i possibili aspetti sanitari e gli effetti sull'organismo umano giocano un ruolo essenziale nel contesto della ricerca sul grafene a livello europeo.<\/p>\n<p>Queste attivit\u00e0 hanno ora dato origine a un ulteriore progetto finanziato dal Fondo Nazionale Svizzero per la Scienza (FNS), che \u00e8 stato recentemente lanciato dall'Empa e dall'AMI. Viene utilizzato un modello di polmone 3-D cellulare, con il quale i ricercatori vogliono scoprire quali effetti il grafene e i materiali simili al grafene possono avere sul polmone umano - e in condizioni che siano il pi\u00f9 possibile vicine alla realt\u00e0. Una sfida, perch\u00e9 non tutto il grafene \u00e8 uguale. A seconda del metodo di produzione e della lavorazione, si produce una grande variet\u00e0 di forme e qualit\u00e0 del materiale, che a sua volta pu\u00f2 scatenare diverse reazioni nei polmoni.<\/p>\n<\/div>\n<\/div>\n<div class=\"std-paragraph-block\">\n<h5>Colture cellulari tridimensionali \"respirano\" le particelle<\/h5>\n<\/div>\n<div class=\"std-paragraph-block\">\n<div class=\"std-art-paragraph\">\n<p>Il team di ricerca guidato da Peter Wick, Tina B\u00fcrki e Jing Wang dell'Empa e Barbara Rothen-Rutishauser e Barbara Drasler dell'AMI ha recentemente pubblicato i primi risultati nella rivista scientifica \"Carbon\". Con il modello polmonare 3-D, i ricercatori sono riusciti a ricreare realisticamente le condizioni reali della barriera aria-sangue e l'effetto del grafene nel tessuto polmonare - senza esperimenti su animali o umani. Si tratta di un modello cellulare che raffigura gli alveoli polmonari. I test convenzionali in vitro lavorano con colture cellulari di un solo tipo di cellula - il modello polmonare stabilito, invece, consiste di tre diversi tipi di cellule che simulano le condizioni all'interno del polmone, cio\u00e8 cellule epiteliali alveolari e due tipi di cellule immunitarie - macrofagi e cellule dendritiche.<\/p>\n<p>Un altro fattore che \u00e8 stato poco considerato finora negli esperimenti in vitro \u00e8 il contatto delle particelle di grafene attraverso l'aria. Di solito, le cellule vengono coltivate in un piatto di coltura in una soluzione nutritiva ed esposte a materiali, per esempio il grafene, in questa forma. Nella realt\u00e0, per\u00f2, cio\u00e8 alla barriera polmonare, \u00e8 diverso.<\/p>\n<p>\"\u00c8 molto probabile che l'organismo umano entri in contatto con le particelle di grafene attraverso l'aria che respiriamo\", dice Tina B\u00fcrki del dipartimento di ricerca Particles-Biology Interactions dell'Empa. Le particelle vengono quindi inalate ed entrano in contatto diretto con il tessuto polmonare. Il nuovo modello di polmone \u00e8 impostato in modo tale che le cellule si trovano su una membrana filtrante porosa all'interfaccia aria-liquido e i ricercatori spruzzano le particelle di grafene sulle cellule polmonari con l'aiuto di un atomizzatore per ricreare il processo nel corpo il pi\u00f9 vicino possibile. La coltura cellulare tridimensionale virtualmente \"respira\" le polveri di grafene.<\/p>\n<\/div>\n<\/div>\n<div class=\"std-paragraph-block\">\n<h5>Nessun danno acuto rilevato<\/h5>\n<p>Questi esperimenti nel modello di polmone 3-D hanno dato i primi risultati. I ricercatori sono stati in grado di dimostrare che nessun danno acuto si forma nel polmone quando le cellule epiteliali del polmone entrano in contatto con l'ossido di grafene (GO) o le cosiddette nanopiastrine di grafene (GNP). Questo include reazioni come la morte cellulare improvvisa, lo stress ossidativo o l'infiammazione.<\/p>\n<div><\/div>\n<div class=\"std-art-paragraph\">Al fine di rilevare anche i cambiamenti cronici nel corpo, il progetto SNSF durer\u00e0 tre anni; il prossimo passo saranno studi a lungo termine con il modello polmonare.
